We\u2019re taking a break for the holidays and will be back in your inbox bright and early on Jan. 3.<\/p>\n<div class=\"ad-in-article dfp-ad-midbreaker-wrap\">\n<p class=\"ad-label\">advertisement<\/p>\n<\/div>\n<h2>The need-to-know this morning<\/h2>\n<ul>\n<li><strong>Bristol Myers Squibb<\/strong>&nbsp;is&nbsp;<a rel=\"noopener\" target=\"_blank\" href=\"https:\/\/www.statnews.com\/2023\/12\/22\/bristol-myers-to-purchase-karuna-maker-of-schizophrenia-drug-for-14-billion\/?utm_campaign=the_readout&amp;utm_source=hs_email&amp;utm_medium=email&amp;_hsenc=p2ANqtz---sPjV33iH13NY3Mr2PvoEJwfuTFXGQpZH12CD_9SNDX55JnvbsB56XjEhlmzGRVoCA1e0\">purchasing<\/a>&nbsp;<strong>Karuna Therapeutics<\/strong>, developer of a drug for schizophrenia, for $14 billion.<\/li>\n<li>The FDA&nbsp;<a rel=\"noopener\" target=\"_blank\" href=\"https:\/\/www.statnews.com\/2023\/12\/21\/fda-approves-astrazeneca-ionis-treatment-for-rare-nerve-disease\/?utm_campaign=the_readout&amp;utm_source=hs_email&amp;utm_medium=email&amp;_hsenc=p2ANqtz---sPjV33iH13NY3Mr2PvoEJwfuTFXGQpZH12CD_9SNDX55JnvbsB56XjEhlmzGRVoCA1e0\">approved a novel medicine<\/a>&nbsp;for a rare and devastating nerve disease called ATTR-PN, clearing a treatment that promises to be more convenient than available therapies. The new drug, called Wainua, is made by&nbsp;AstraZeneca&nbsp;and&nbsp;Ionis Pharmaceuticals.<\/li>\n<li><strong>Allovir<\/strong>&nbsp;is&nbsp;<a rel=\"noopener\" target=\"_blank\" href=\"https:\/\/ir.allovir.com\/news-releases\/news-release-details\/allovir-provides-updates-phase-3-clinical-development-program?utm_campaign=the_readout&amp;utm_source=hs_email&amp;utm_medium=email&amp;_hsenc=p2ANqtz---sPjV33iH13NY3Mr2PvoEJwfuTFXGQpZH12CD_9SNDX55JnvbsB56XjEhlmzGRVoCA1e0\" data-hs-link-id=\"0\">shutting down Phase 3 clinical trials<\/a>&nbsp;of an off-the-shelf T-cell therapy for three types of infectious disease. The decision comes after independent study monitors concluded Allovir\u2019s therapy was unlikely to achieve its efficacy goals.<\/li>\n<li>The FDA prescribing label for Carvykti, a CAR-T therapy for multiple myeloma made by&nbsp;Johnson &amp; Johnson&nbsp;and&nbsp;Legend Biotech,&nbsp;<a rel=\"noopener\" target=\"_blank\" href=\"https:\/\/www.sec.gov\/Archives\/edgar\/data\/1801198\/000115752323001857\/a53874140.htm?utm_campaign=the_readout&amp;utm_source=hs_email&amp;utm_medium=email&amp;_hsenc=p2ANqtz---sPjV33iH13NY3Mr2PvoEJwfuTFXGQpZH12CD_9SNDX55JnvbsB56XjEhlmzGRVoCA1e0\" data-hs-link-id=\"0\">has been updated<\/a>&nbsp;to include the occurrence of secondary blood cancers in patients treated with the medication.<\/li>\n<li><strong>Sarepta Therapeutics<\/strong>&nbsp;said an&nbsp;<a rel=\"noopener\" target=\"_blank\" href=\"https:\/\/investorrelations.sarepta.com\/news-releases\/news-release-details\/sarepta-therapeutics-submits-efficacy-supplement-expand-elevidys?utm_campaign=the_readout&amp;utm_source=hs_email&amp;utm_medium=email&amp;_hsenc=p2ANqtz---sPjV33iH13NY3Mr2PvoEJwfuTFXGQpZH12CD_9SNDX55JnvbsB56XjEhlmzGRVoCA1e0\" data-hs-link-id=\"0\">efficacy supplement was submitted<\/a> to the FDA, seeking to expand the prescribing label of its gene therapy for Duchenne muscular dystrophy to all patients without restrictions on age or ambulatory status.<\/li>\n<\/ul>\n<h2>Biotech\u2019s green 2023 is in doubt<\/h2>\n<p>After an extended bull run that had investors looking rosily forward to 2024, the closely watched XBI biotech index had its worst day since February on Wednesday, falling more than 4%. Then it recovered most of those losses yesterday in another dramatic move that didn\u2019t appear tied to the actual fundamentals of developing new medicines, much less a single company\u2019s misfortunes.<\/p>\n<p>Biotech seems to be heading to a volatile end of what has been a volatile year, in which the sector has been up as much as 13% and down by more than 20%. And while the biotech\u2019s recent recovery is clearly tied to macroeconomic factors, Wednesday\u2019s sudden dip offers few easy explanations beyond investors taking profits, year-end tax-loss selling, and the familiar animal spirits.<\/p>\n<p>The XBI is now up about 4% on the year, which is better than 2022 but far worse than the S&amp;P 500\u2019s 24% return since January. Investors seem to believe things are going to look better in 2024. Mizuho surveyed 70 fund managers and found that 71% expect biotech to outperform the S&amp;P next year, with 11% expecting the sector to underperform and 17% figuring it will be in-line with the broader market.<\/p>\n<div class=\"ad-in-article dfp-ad-midbreaker2-wrap\">\n<p class=\"ad-label\">advertisement<\/p>\n<\/div>\n<h2>Can biotech endure another election year?<\/h2>\n<p>What does it mean to a bad CEO? It\u2019s our last episode of 2023, so we look back on the biggest stories of the year, discuss the best and worst CEOs in the industry, and make some reasonably informed guesses on what 2024 has in store for the drug industry.<\/p>\n<p><a rel=\"noopener\" target=\"_blank\" href=\"https:\/\/www.statnews.com\/2023\/12\/21\/readout-loud-podcast-2023-recap-biotech-ceos\/?utm_campaign=the_readout&amp;utm_source=hs_email&amp;utm_medium=email&amp;_hsenc=p2ANqtz---sPjV33iH13NY3Mr2PvoEJwfuTFXGQpZH12CD_9SNDX55JnvbsB56XjEhlmzGRVoCA1e0\">Listen here.<\/a><\/p>\n<h2>Cytokinetics is running out of December<\/h2>\n<p>The last big biotech catalyst for 2023 belongs to Cytokinetics, which has&nbsp;<a rel=\"noopener\" target=\"_blank\" href=\"https:\/\/www.statnews.com\/2023\/02\/27\/for-cytokinetics-an-fda-rejection-of-its-heart-drug-would-help-not-hinder\/?utm_campaign=the_readout&amp;utm_source=hs_email&amp;utm_medium=email&amp;_hsenc=p2ANqtz---sPjV33iH13NY3Mr2PvoEJwfuTFXGQpZH12CD_9SNDX55JnvbsB56XjEhlmzGRVoCA1e0\">spent more than 25 years working toward a first FDA approval<\/a>, and the clock is ticking.<\/p>\n<p>The company has promised Phase 3 data that will determine whether its drug, aficamten, can improve the lives of patients with a type of inherited heart disease. Those results are due in \u201clate December,\u201d the company has said, which means they should be here any day now. Investors seem to think the news is going to be good, as Cytokinetics stock price rose 25% yesterday.<\/p>\n<p>The stakes are high. If aficamten performs better than&nbsp;<a rel=\"noopener\" target=\"_blank\" href=\"https:\/\/www.statnews.com\/2020\/10\/05\/bristol-myers-squibb-buy-myokardia-for-13-billion\/?utm_campaign=the_readout&amp;utm_source=hs_email&amp;utm_medium=email&amp;_hsenc=p2ANqtz---sPjV33iH13NY3Mr2PvoEJwfuTFXGQpZH12CD_9SNDX55JnvbsB56XjEhlmzGRVoCA1e0\">a rival drug from Bristol Myers Squibb<\/a>, Cytokinetics could roughly double in value. If the drug simply measures up to its competition, the company will still be on pace to launch its first commercial product. Meanwhile, Cytokinetics has&nbsp;<a rel=\"noopener\" target=\"_blank\" href=\"https:\/\/www.bloomberg.com\/news\/articles\/2023-12-21\/biotech-cytokinetics-jumps-after-report-of-takeover-interest?utm_campaign=the_readout&amp;utm_source=hs_email&amp;utm_medium=email&amp;_hsenc=p2ANqtz---sPjV33iH13NY3Mr2PvoEJwfuTFXGQpZH12CD_9SNDX55JnvbsB56XjEhlmzGRVoCA1e0\" data-hs-link-id=\"0\">reportedly attracted buyout interest from larger drugmakers<\/a>, a prospect that is almost certainly contingent on aficamten\u2019s success.<\/p>\n<h2>Pharma still can\u2019t figure out PTSD<\/h2>\n<p>Despite years of research and a few promising advances, there remain only two approved treatments for post-traumatic stress disorder, a pair of now-generic SSRIs drugs. And the pharmaceutical industry\u2019s latest idea, an investigational treatment from Jazz Pharmaceuticals, ended in failure.<\/p>\n<p>The drug, JZP150, missed its primary and secondary endpoints in a Phase 2 trial,&nbsp;<a rel=\"noopener\" target=\"_blank\" href=\"https:\/\/www.prnewswire.com\/news-releases\/jazz-pharmaceuticals-provides-update-on-phase-2-trial-of-investigational-jzp150-in-adult-patients-with-post-traumatic-stress-disorder-302021299.html?utm_campaign=the_readout&amp;utm_source=hs_email&amp;utm_medium=email&amp;_hsenc=p2ANqtz---sPjV33iH13NY3Mr2PvoEJwfuTFXGQpZH12CD_9SNDX55JnvbsB56XjEhlmzGRVoCA1e0\" data-hs-link-id=\"0\">the company said yesterday<\/a>, failing to meaningfully improve symptoms of PTSD relative to placebo over the course of a 12-week study. Jazz said it does not expect to conduct further trials of JZP150 in PTSD.<\/p>\n<p>The most promising pipeline medicines for PTSD&nbsp;<a rel=\"noopener\" target=\"_blank\" href=\"https:\/\/www.statnews.com\/2023\/06\/29\/psychedelics-ptsd-maps-wrestles-with-identity\/?utm_campaign=the_readout&amp;utm_source=hs_email&amp;utm_medium=email&amp;_hsenc=p2ANqtz---sPjV33iH13NY3Mr2PvoEJwfuTFXGQpZH12CD_9SNDX55JnvbsB56XjEhlmzGRVoCA1e0\">come from the world of psychedelics<\/a>.
